My first suggestion would be the firewall client be updated as the original (with ISA2000) won't run on Vista. Assume you visited http://www.microsoft.com/downloads/details.aspx?FamilyID=05C2C932-B15A-4990-B525-66380743DA89&displaylang=en and got the latest client. Do any other Vista machines work OK? ie: is the problem limited to settings on just one client machine? Beyond that, can the client access an internal SMTP or POP3 server? ie: Are you sure it's not client level firewall stopping the access? Lastly, logs are your friends. Check out the firewall client logs (and securenat clietn logs if appropriate) on the ISA server to see what access is being denied. Are you certain rulesets allow this? Is this a "new" problem that has just started, or has it alwways been the case that Vista machines couldnt access external POP3/SMTP? If a "new" problem, what changed most recently?

Can the client machine resolve the name of the exernal server back to an IP? If not, troubleshoot DNS issues. Can the client connect to an INTERNAL mail server on 25/110 for SMTP/POP3? If not, it's definitely a "client side" issue so look at firewall settings on the client.. or perhaps QOS may be screwing something up. Lastly, the logs will be able to identify what, if anything is specfically denying or at least not dispaly the rule your are expcting to GIVE access which you can then troubleshoot.

every other client connects out OK vista machine can resolve dns name of mail server OK access from vista machine still denied. What's the common link here??? 3 minute job to enable a SMTP listener on an internal ISS machine which should verify no firewall on the Vista box is stopping the smtp/pop3. Beyond that....? shrug.
