ISA Capacity/Scailability

I'm a bit of a novice for ISA.  I have deployed ISA 2006 only in Standard.  What I'm curious about, is scailability for Exchange reverse proxy publishing.

Let's say we have 5,000 users.  We're going to deploy ISA.  What are our options?  Even let's say we had 50,000 users. 

1.  Would an array be enough?
2.  Would we want to use NLB inside our array?  Why use NLB in our array?  What benefits does NLB give us over using no NLB within our array?
3.  For scalability for let's say 50,000 users, would we use multiple arrays that use NLB within that array and then use a hardware load balancer to distribute the load between the different arrays with NLB enabled within them?

How does this really work?
