I have an integrated-mode ISA box providing transparent caching for a subnet in my lab. The clients on this subnet use a specific TCP port (7778) to access an in-house application. The application contains rather large TIFF files as part of its data set and I would like the ISA box to cache these TIFF files. Is there a way to get ISA Server to cache content based on TCP port number?

Not that I know of. However, you could configure your DNS or the ISA Server to "loop back" to obtain content on the internal network. This generally is highly un-recommended, but if you have a compelling reason to cache these large files, it could be done. It will result in generally decreased performance for rest of your Internet access, though.
